No Offer Description Applications are open for one Post-Doctoral Research Fellowship, within the framework of "A Gold Nanosensor for Single-Molecule Detection of RNA Biomarkers in a Miniaturized Microscope" / CQE, IST-ID, Ref. 2022.04076.PTDC, financed by national funds through FCT/MCTES (PIDDAC). Workplan: Porphyrins are versatile optically active molecules, which have potential as luminescent labels with emission in the near infrared. In this work plan, the hired researcher will perform the synthesis of novel water-soluble sugar-substituted porphyrin derivatives using in-house developed methods. The porphyrins will be derivatized with oligonucleotide sequences for their incorporation into optical sensors for nucleic acid detection. The phosphorescence emission from porphyrin dyes will be intensified by optical nanogap antennas to achieve better sensitivity in readout signals. The detailed objectives are: Synthesis of adequate porphyrins to be used as the core platform for different biomoieties, such as carbohydrates. Metalation of the above glycoporphyrins with Zn(II), Mn(II), Pd(II) or Cd(II) in order to promote spin-orbit coupling and high phosphorescence quantum yields. Include custom-synthesized DNA oligonucleotides in the prepared porphyrin complexes. Structural and photophysical characterization of all the intermediates and final products. Duration: The research fellowship will have the duration of 12 months. It's expected to begin in October 2024 and may be eventually renewed.
