Offer Description SUMOylation and ubiquitination are two types of protein post-translational modifications (PTMs) that control virtually all physiological and pathophysiological processes. SUMO and ubiquitin E3 ligases have been implicated in several types of cancer. However, although the changes in these PTMs have been clearly correlated with the initiation and progression of cancer, very little is known about the molecular mechanisms involved in these processes and the role of E3 ligases. Nevertheless, a growing body of work published in recent years highlights the key role of SUMOylation and ubiquitination enzymes in cancer, and the possibility of using them as diagnostic and/or prognostic biomarkers to predict patient response to anti-tumoral treatments and survival.
The prospective post-doc's research project will focus mainly on the biochemical and structural characterisation of selected E3 ligases and their substrates, in particular the nucleosome/chromatin.
Key Responsibilities:
Biochemistry and molecular biology techniques, protein and DNA purification, cell biology.
Preparation of samples for electron-microscopy and cryo-EM, observation and analysis of DNA-protein complexes and chromatin.
Interpretation of results and writing of articles.
Presentation of results at conferences and scientific events.
The CBM (UPR4301-CNRS) is located on the CNRS campus in Orléans, which has 6,300m2 of modern, functional buildings. It is home to biologists, physicists, and chemists who work in close collaboration to understand the molecular mechanisms that lead to the development of certain diseases such as cancer. The research centre provides an interactive and international working environment, with a wide range of instruments suitable for multidisciplinary research, from cellular and preclinical imaging to crystallography. This makes it possible to work in an environment conducive to exchanges with top scientists specialising in complementary fields. The project is part of a larger ERC-funded project on protein SUMOylation entitled "SUMOwriteNread" (grant id 101078837).
The postdoc will be under the scientific guidance of Dr Marcin SUSKIEWICZ and Dr Ibtissam TALHAOUI, within the "Protein post-translational modifications and DNA repair: structure, function, and dynamics" dynamic interdisciplinary team, co-led by Dr Bertrand CASTAING and Dr Marcin SUSKIEWICZ.

Additional Information Eligibility criteria:
We are looking for a motivated, dynamic, experienced, and independent postdoctoral researcher. The successful candidate will ideally have solid experience in the study of post-translational modifications of proteins, in particular SUMOylation and ubiquitination. S/he should have solid expertise in in vitro chromatin reconstitution. Experience in electron microscopy (EM) and/or cryo-EM would be appreciated but not essential. Knowledge of biochemistry and molecular biology techniques is required. Previous experience in cell biology would be an advantage but is not essential. S/he should be able to carry out his/her research project independently and work closely with other team members. S/he should also be able to collaborate with external laboratories. S/he will benefit from our support in applying for postdoctoral fellowships in order to continue working on this scientific project beyond the originally planned period and to pursue his/her scientific career.
