The Inserm UMR-1250 P3Cell unit in Reims is a partner in the ANR-22-CE44-0024 project "IRon Uptake Pathways in Pseudomonas aeruginosa : phenotypic switches and mechanism of regulation- IRUPP".
One of the main obstacles to the development of new antibiotics is the ability of active molecules to penetrate bacterial cells. Iron import pathways are often proposed to import antibiotics into bacteria, thus circumventing the low permeability of the bacterial membrane. To gain access to iron, an essential nutrient, bacteria synthesize siderophores. Siderophores are released into the bacterial environment, where they capture iron before being reabsorbed by the bacteria. Antibiotics can be attached to siderophores, so when the pathogen absorbs the ferri-siderophore, it imports the antibiotic.
As part of IRUPP, the Inserm UMR-1250 P3Cell unit aims to study the plasticity of expression of different iron uptake pathways during infection of the human airway epithelium by P. aeruginosa .
Profile: Research engineer, 1 year renewable, starting January 1, 2025

6. Principal Activities Design the development and carry out the specialist testing of bacterial infections of human airway epithelia obtained from primary cells cultured in an air-liquid interface.
Bacterial cultures to prepare infecting bacterial inocula (L2).
Primary cell cultures to obtain the ex vivo model of human airway epithelium.
Experimental infection monitoring and time-lapse microscopy.
Infectious waste management.
Advise on technical options within the framework of the project; evaluate and validate experimental choices.
